第一次做多态性位点的STR全自动分型，是送生工做的，结果生工只返回了PDF的图片和excel数据，我看不懂，但生工没有技术支持，已经一周了，没有任何回音。特上DXY请教：
1、表格中的数据分别是什么含义？

2、图中实心峰（图标1、2）是否就是测得的多态位点？
3、图中紧挨实心峰的空心峰（图标3）是什么？
4、图中小的空心峰（图标4）是什么含义？
5、图中实心峰2远大于实心峰1，若峰值两倍于1，能否判定1处为1个STR位点，2处为2个STR位点，即整个结果可以判定为三体或该位点处为三体？

建议用bin设好基因型也就是取整
1、明显看到上样样本浓度过高
已经超过机器读取最高界，建议稀释后
2、1-4全是杂峰，2-3是主峰，但具体怎么
判断，要把峰都降下来。

是直接将PCR产物的浓度稀释，还是将DNA标本浓度稀释后重新PCR？

生工的结果就不咋样，二倍体物种一般都要给出两个alleles的峰值，峰高，峰面积，以便判断杂合还是纯合子。1.2.3的峰值没给出则无法判断哪个是真实的峰图，4为杂峰。
ps:坛里看到STR峰图判读的帖子太少了，欢迎有经验的朋友来交流

1、上样浓度过高，同意至少做10倍稀释再上样；
2、genemapper是AB公司的商业软件，价格昂贵，没有免费下载地址；
3、做SSR分析时，最好不要选用单核苷酸（如polyA、polyG等）、二核苷酸（如AT、AG等的重复）重复的SSR位点，至少要选用3核苷酸重复的SSR位点，最好是4或5核苷酸重复的SSR位点，否则影子带太严重，会严重干扰分析。
4、峰2和峰3相差大约1个碱基，可能的结果是：峰3是峰2的n+1峰，而其余的峰相对于峰2/3而言太低，可能是杂峰。具体的结果只有等样品稀释后，峰2/3的结果完整后才能判断。另，判断是主峰还是杂峰，不是以峰高为标准，更重要的是以峰面积为判断标准。The page is temporarily unavailable
